When you purchase a Kindle 2 is it possible to request specific titles that may not be in the Amazon kindle store?

No, you cannot. Actually, Amazon want to have all books available in Kindle format because it can further boost up the sales of their Kindle. It is actually the publisher / author who decide whether they want their books to publish in Kindle format.
There is a link "I’d like to read this book on Kindle" on the left hand side on the sales page of every book (like the one in the link below). Every one can click it and when there are enough clicks it usually works:

BTW, the number of books available for the Kindle is keep on increasing. I remember there are 270,000 Kindle books when I bought my Kindle 2 a few months ago, but now there are over 340,000 books, as you can see here:

You cannot. But on every paper book available on Amazon, there is a link that you can tell the publisher you want the book in Kindle format.

No, you cannot. But actually more and more books are available for the Kindle. You may find a book not available a month ago but now available. You can always do a quick search here among the list of all available Kindle books:
